چکیده

cell ghosts (Steck et. al., Science 168:255(1970)). Lipid membranes, the sheets which form the boundaries of cells, must maintain their integrity for cells to live [1]. Since a tear in a membrane exposes the hydrophobic interior of the sheet to water, tradition maintains that membranes have a high line tension (energy per unit length of exposed edge) and never spontaneously form holes. We show that membranes can form holes in a simple model of a charged membrane. By examining the competition between electrostatic repulsion and line tension, we find a parameter range where electrostatics dominates and membranes are unstable to hole formation. Although much previous work examines electrostatic effects on lipid assemblies [2] and the bending moduli of membranes [3–5], we are only aware of one study addressing electrostatics and line tension [6].

Elasticity and mechanical instability of charged lipid bilayers in ionic solutions.

We use coarse-grained Monte Carlo simulations to study the elastic properties of charged membranes in solutions of monovalent and pentavalent counterions. The simulation results of the two cases reveal trends opposite to each other. The bending rigidity and projected area increase with the membrane charge density for monovalent counterions, while they decrease for the pentavalent ions. These ob...

Interaction forces between ternary lipid bilayers containing cholesterol.

Interaction force-distance profiles between substrate-supported membranes composed of equimolar ternary mixtures of unsaturated phosphotidylcholine (PC) lipid, saturated PC lipid, and cholesterol were determined using the surface force apparatus. Both double and single unsaturated PC lipids were studied. In all cases, the membranes were slightly negatively charged, resulting in a weak, long-ran...

Electrostatic interaction effects on tension-induced pore formation in lipid membranes.

We investigated the effects of electrostatic interactions on the rate constant (k(p)) for tension-induced pore formation in lipid membranes of giant unilamellar vesicles under constant applied tension. A decrease in salt concentration in solution as well as an increase in surface charge density of the membranes increased k(p). These data indicate that k(p) increases as the extent of electrostat...

The effect of lipid demixing on the electrostatic interaction of planar membranes across a salt solution.

We study the effect of lipid demixing on the electrostatic interaction of two oppositely-charged membranes in solution, modeled here as an incompressible two-dimensional fluid mixture of neutral and charged mobile lipids. We calculate, within linear and nonlinear Poisson-Boltzmann theory, the membrane separation at which the net electrostatic force between the membranes vanishes, for a variety ...

Binding of imipramine to phospholipid bilayers using radioligand binding assay.

Binding of the tricyclic antidepressant imipramine (IMI) to neutral and negatively charged lipid membranes was investigated using a radioligand binding assay combined with centrifugation or filtration. Lipid bilayers were composed of brain phosphatidylcholine (PC) and phosphatidylserine (PS). IMI binding isotherms were measured up to IMI concentration of 0.5 mmol/l. Due to electrostatic attract...
